You cannot specify the book as you have "is" before it. You can tell a person that there has been 'a' book you are looking for all day long. You cannot specify it as the other person does not know the book. You are singling them that you have been looking for some book all day. You cannot specify the book due to the presence of the verb 'is'. But you can say 'this is the book I have been looking for all day.'
